DNA Extraction from Trophozoites and Cysts Using Manual Protocol

Protist taxa are ubiquitous members among soil microbial communities. Soil protistan communities are very diverse and yet to be explored due to the complex isolation and separation procedure. They have two forms in their life cycle: trophozoites and cyst. However, isolation of protistan DNA is much difficult due to insufficient cell lysis. Many commercial and advanced kit-based methods are available for DNA extraction, but the best one depends on high quality and greatest yield.
