Durable response to cancer therapies relies on the stimulation of cancer immunosurveillance. Reinstating antitumor immunity can be achieved by the administration of cytotoxic treatments able to induce a nontolerogenic type of cell death. This so-called immunogenic cell death (ICD) is characterized by the exposure or release of damage-associated molecular patterns, the subsequent recruitment and activation of antigen-presenting cells, and the ultimate priming of tumor-targeting T lymphocytes. The present paper will recapitulate the cellular features of ICD and reports both intrinsic and extrinsic determinants modulating this phenomenon. Therapeutic agents able to trigger this malignant cell demise are listed and the methodological approaches allowing their screening are documented. At last, clues supporting the clinical occurrence of ICD and its interest in sensitizing tumors to immunotherapies are highlighted.
