Polyphenolic-rich extract of Phyllanthus niruri (Linn.) improves antioxidant status and modulates Akt/Nrf2 and IR/Akt/GLUT2 pathways in streptozotocin-induced diabetic rats

Medicinal plants have proven to be invaluable in the management of diabetes, whose prevalence has continued to increase in several parts of the world. This study evaluated the ameliorative effect of polyphenolic-rich extract of Phyllanthus niruri (Linn.) (PEPN) on antioxidant status and glucose metabolism in streptozotocin-induced Wistar rats.
MethodsIn vivo antidiabetic study was conducted using fifty-six Wistar rats that were randomly distributed into 7 groups with eight rats each (n = 8) including normal control, diabetic untreated as well as diabetic rats treated with 5/500 mg/kg bw glibenclamide/metformin and various doses of PEPN. Activities of catalase and glutathione peroxidase, as well as levels of malondialdehyde were evaluated in the liver. Hepatic activity and level of hexokinase and glycogen were also respectively assessed. Expression of insulin receptor (IR), glucose transporter-2 (GLUT2) and Akt were also monitored.
ResultsPEPN demonstrated appreciable in vitro antioxidant activity and inhibited α-amylase and α-glucosidase in all the concentrations tested. Likewise, PEPN at the highest dose considered (i.e., 125 mg/kg), improved antioxidant status of diabetic rats and elevated hexokinase activity and glycogen levels. Also, PEPN upregulated the expression of IR, Akt at 75 mg/kg, while GLUT2 and Nrf2, mostly at least dose of 50 mg/kg.
ConclusionOur results indicate that PEPN exerts its antidiabetic activity via improving cellular antioxidant capacity and glucose metabolism, and that the plant could be relevant in the management of diabetes.
