Cysteine improved healing effects of cimetidine on acetic acid-induced gastric ulcer in male Wistar rats

Gastric ulcers, defined by disruptions in the stomach lining, pose significant global health challenges, leading to considerable morbidity and mortality. Cimetidine, a common treatment for acute gastric ulcers, only mitigates acid secretion and does not fully address NSAID-induced damage while Cysteine, an amino acid offers potential antioxidant and vasodilatory properties and may provide additional benefits. Hence, this study investigates the combined effects of cysteine and cimetidine on acetic acid-induced gastric ulcers and explores their mechanisms of action.
MethodsSeventy-five rats (140 ± 10 g) were randomly allocated into five groups (n = 15). Group I = (positive no ulcer control group). Gastric ulcers were induced in Group II-V via 0.2 ml acetic acid injection. Group II = negative ulcer group and received no treatment after ulcer induction. Groups III = cysteine alone (300 mg/kg), Group IV = cimetidine alone (50 mg/kg), and Group V = combination of cystiene and cimetidine. Rats were sacrificed on days 3, 7, and 14 for analysis. Blood samples were assessed for SOD activity, NO levels, and MDA concentration. TAC, Inflammatory markers (TNF-α, IL-1β, and IL-10) were also assayed for. Data were analyzed using ANOVA (P < 0.05).
ResultsThe ulcer score was significantly reduced in the cysteine (0.36 mm2), cimetidine (0.67 mm2), and combination (0.28 mm2) groups compared to the ulcer group (1.366 mm2), with percentage inhibitions of 73.2%, 50.91%, and 85.3%, respectively. Higher SOD activity, NO levels, and TAC, along with lower MDA, TNF-α, IL-1β, and IL-10 levels, were observed with the combined treatment.
ConclusionL-cysteine enhances the healing effects of cimetidine in acetic acid-induced gastric ulcers by boosting antioxidant defenses and reducing inflammation.
