<p>Tropomyosin is one of the key components involved in actin–myosin interactions, which are essential for muscle contraction and regulation of cytoskeletal dynamics. While well-established in striated muscle, this study reinforces the presence and structural organization of tropomyosin within the smooth muscle thin filament, utilizing advanced techniques like negative staining and structural analysis using the IHRSR method to provide further clarity on its arrangement. Thin filaments prepared from chicken gizzard were analyzed by SDS-PAGE, revealing two distinct bands corresponding to actin and tropomyosin, with the identity of tropomyosin further confirmed by Western blotting. Structural analysis using negative staining transmission electron microscopy (TEM) provided critical insights into the organization of tropomyosin within the thin filament under calcium-depleted conditions. Confirmation of tropomyosin’s integration within the smooth muscle thin filament provides significant structural context for its functional role and encourages further investigation into its physiological implications.</p>
