Identification and expression analysis of apple CYP family genes in different rootstock-scion combination

To investigate the relationship between Cytochrome P450s (CYP450) gene family members and plant hormones on scion growth of different rootstocks. Through the determination of hormone content in leaves of different anvil groups, 359 members of apple CYP450 family were identified by Pfam number of CYP450, and their expression patterns were analyzed by bioinformatics. The results showed that the contents of IAA and GA3 were highest in Ha + Ms, but lower in Ha + G935. Bioinformatics analysis showed that 359 family members were distributed on 17 chromosomes, encoding 65 ~ 1653 amino acids, molecular weight of 7.3 ~ 188.4 kD, isoelectric point of 4.58 ~ 10.91. The gene family can be divided into five subfamilies with exon numbers ranging from 1 to 14. Selection pressure analysis showed that 82 pairs of genes were subjected to purification selection during evolution. Real-time fluorescence quantitative PCR showed that the expression levels of MdCYP33 and MdCYP126 in Ha + T337 were more than 10 times that of Ha + Ms. The expression of MdCY355 in Ha + QZ1 is more than 20 times that of Ha + Ms, and these findings reveal the potential importance of the CYPs gene in highly regulated plants.
