Abstract <p><b>Objectives:</b> The aim of this study was to investigate the mechanisms of anti-inflammatory effect of oleoylethanolamide (OEA)-based dietary supplement (OEA-DS) in diet-induced obesity in mice. The degree of inflammatory response in omentum tissue was evaluated by immunohistochemical study of iba-1, CD68 and CD163 markers. <b>Methods:</b> In vitro, OEA-DS-dependent changes in the synthesis of the cytokine IL-10, as well as the markers CD163 and CD206 were studied on macrophage cell cultures by Western blotting. The regularities of changes in PPAR-α activation under the action of OEA-DS were studied in omentum tissue in vivo and in macrophage cell culture in vitro. <b>Results:</b> It was found that OEA-DS administration in obesity leads not only to a decrease in inflammatory response (iba-1) in visceral adipose tissue, but also to suppression of proinflammatory (CD68) and stimulation of anti-inflammatory (CD163) type of macrophage activation. In addition, OEA-DS was found to stimulate the production of anti-inflammatory markers such as IL-10, CD206 and CD163 in macrophage cell culture in vitro. <b>Conclusion:</b> Thus, the complex anti-inflammatory effect of OEA-DS together with anorexigenic effects allows it to influence several pathogenetic links of obesity at once.</p>
