Abstract <p><b>Objectives:</b> In this study, the viability and adhesive properties of BRO and SK-MEL-2 melanoma cells were assessed. <b>Materials and methods:</b> It was shown that in BRO cells, apoptosis after exposure to dacarbazine was combined with the transition of a proportion of cells into the G<sub>0</sub> phase of the cell cycle, which confirms the previously obtained results. In SK-MEL-2 cells, no apoptosis and no exit from the cell cycle were observed in the 3D spheroids. It was also revealed that in control spheroids (cells without drug exposure) of BRO and SK-MEL-2 melanoma cell lines, adhesion to fibronectin was higher compared to control monolayer cells, which is explained by the three-dimensional structure requiring cell communication with the extracellular matrix. <b>Results and discussion:</b> In spheroids formed by SK-MEL-2 cells, dacarbazine induced a decrease in adhesion to fibronectin, which may be associated with the development of drug resistance. <b>Conclusion:</b> After exposure to dacarbazine, the expression of integrins αV and β8 in BRO and SK-MEL-2 cells, as well as of integrin β5 in SK-MEL-2 cells were increased, which may indicate the involvement of these molecules in the loss of the proliferative status of tumor cells.</p>
