Abstract <p>A combination of lithium chloride and organic sulfonic acid was used to efficiently remove <i>O</i>-methyl groups to prepare dotinurad and its analogues, and the feasibility of the method was verified through kilogram-level experiments. Detection methods for dotinurad and its related substances were developed for quality control purposes.</p>
