Construction of Fisher Information Matrix for Multiparametric Problem in Super-Resolution Optical Fluctuation Imaging

Abstract
We propose a method of Fisher information calculation for super-resolution optical fluctuation imaging (SOFI) with a realistic multiparametric model, taking into account finite acquisition time and non-trivial emitters’ blinking induced signal correlations. Computational efficiency is ensured by separation of geometrical effects from statistical description of emitters’ blinking and shot noise. Fisher information matrices were successfully constructed for SOFI images up to the 4th order and used for spatial resolution quantification.
