Cytotoxic, Antioxidant, and Antimicrobial Activities of Inula graveolens and Inula viscosa: Solvent-Dependent Bioactivity and Therapeutic Potential

This study evaluates the cytotoxic, antioxidant, and antimicrobial properties of methanol, acetone, and aqueous extracts of Inula graveolens and Inula viscosa collected in Turkey. Antimicrobial activity was assessed by determining the inhibition zones against two Gram-positive (Staphylococcus aureus ATCC25923, Bacillus subtilis) and two Gram-negative (Escherichia coli ATCC 25922, Pseudomonas aeruginosa ATCC 27853) bacterial strains. Cytotoxic effects against three human cancer cell lines (prostate PC-3, hepatocellular Huh-7, and colorectal Caco-2) were evaluated via the MTT assay. Antioxidant potential was quantified using ABTS radical scavenging and phosphomolybdenum reduction assays. Results demonstrated dose-dependent cytotoxicity with IC50 values ranging from 10.3 ± 0.1 to 190.0 ± 2.9 µg/mL across all cell lines. Aqueous extracts of both species exhibited superior antioxidant activity (IC50: 0.15 ± 0.003 mg/mL for I. graveolens vs. 0.24 ± 0.003 mg/mL for methanol extracts), particularly in I. graveolens. Antimicrobial effects were observed against all strains, with the aqueous extract of I. viscosa showing the highest efficacy against S. aureus (25 mm inhibition zone vs. 22 mm for kanamycin). These findings highlight the potential of Inula graveolens and Inula viscosa as promising sources of natural antimicrobial and anticancer agents, warranting further exploration of their bioactive constituents.
