Abstract <p>Oxytocin (OXT), a hypothalamic neurohormone secreted in the central nervous system and entering the peripheral circulation via the posterior pituitary gland, is implicated in the regulation of various physiological functions including reproductive behavior, metabolism, social behavior, eating behavior, and body weight control. In this study, we investigated the association of <i>OXT</i> and oxytocin hormone receptor (<i>OXTR</i>) gene variants with obesity and food addiction in the Turkish population. The <i>OXT</i> rs2770378, <i>OXTR</i> rs53576, and <i>OXTR</i> rs2254298 variants were analyzed by Polymerase Chain Reaction-Restriction Fragment Length Polymorphism (PCR-RFLP) method in 80 obese and 80 healthy study participants. The frequency of individuals with food addiction was higher in the obese group compared to the control group (<i>p</i> &lt; 0.001). No statistically significant difference in the genotype distribution of the <i>OXT</i> rs2770378 and <i>OXTR</i> rs2254298 variants between the patient and control groups (<i>p</i> = 0.819, <i>p</i> = 0.221, respectively). The <i>OXTR</i> rs53576 AA genotype was significantly higher in the patient group compared to the control group, while the GG genotype was more common in the control group compared to the patient group (<i>p</i> = 0.002). In conclusion, this is the first study demonstrating <i>OXTR</i> rs53576 association with food addiction in the Turkish population. We suggest that the <i>OXTR</i> rs53576 AA genotype may be a risk factor for obesity, whereas the GG genotype may be potentially protective.</p>
