DNA Barcoding and Comparative Bioinformatic Analysis of the Endangered Genus Herpetospermum with Special Reference to Herpetospermum darjeelingense

DNA barcoding technique employs the methodology of identifying species based on minuscule sections of DNA from conserved genes serving as an imperative tool towards decoding taxonomic distinctiveness to anonymous species. An indigenous ethnomedicinal plant of Darjeeling eastern Himalaya named Herpetospermum darjeelingense (C.B. Clarke) H. Schaef. and S.S. Renner has remained immensely unexplored from the point of view of molecular characterization. The genus Herpetospermum constitutes three species viz. H. darjeelingense; Herpetospermum pedunculosum (Ser.) Baill. and Herpetospermum tonglense (C.B. Clarke) H. Schaef. and S.S. Renner with the present research aiming to confer rbcL gene based DNA barcode to H. darjeelingense including a comparative bioinformatic analysis of the rbcL gene data with its allied cucurbit species. Findings of this investigation led to recognizing relatedness of H. darjeelingense to Australian endemic plant Nothoalsomitra suberosa including its Tibetan sister H. pedunculosum and H. tonglense. rbcL gene based similarity of H. darjeelingense to N. suberosa denotes divergent evolutionary approach with supposed co-habilitation of these plants at alike localities archaically being isolated subsequently post splitting of supercontinent Pangaea. Bioinformatic analysis of the genus Herpetospermum revealed considerable variation with conserved identity among its sister species. Prospects of further study with complete genomic dataset could further aid in introspecting the evolutionary lineage of Herpetospermum.
