Genotoxic, Biochemical and Morphotoxic Assessment of Dimethoate: Ameliorative Role of Grape Seed (Vitis vinifera L.) Extract

The chief objective of this investigation has been to identify and evaluate, employing a battery of morphological, cytological, biochemical and molecular bioassays, the mechanism of action of the dimethoate toxicity and the protective role of grape seed extract (GSE) against toxicity on model plant Allium cepa L. IC50 dose was 125 mg/L in dimethoate treatment regime (96 h) producing root growth inhibition, final and fresh root length and root weight of bulbs significantly; in dimethoate + GSE treated Gr-V and VI (i.e. 125 mg/L dimethoate + 75 mg/L GSE, 125 mg/L dimethoate + 150 mg/L GSE) there was the significant dose-dependent revival of all morphological parameters. Predominantly, sticky chromosome, fragments, unequal distribution of chromatin, vagrant chromosome, c-mitosis, binucleated cells and nuclear lesions were seen (Gr-IV), while CAs percentages had been lessened in group V and VI and GSE partially blocked the MN formation in comparison to Gr-IV where dimethoate applied alone. Interestingly, in Gr-IV pronounced damage in genomic DNA was evident, but dimethoate in combination with GSE (group V and VI) remarkably improved DNA tail percentage in the comet assay. Increased MDA, SOD, CAT and GR activity in Gr-IV where dimethoate applied alone was accounted whereas GSE lessened the MDA formation and diminished the dimethoate-mediated attenuation of SOD, CAT and GR (Gr-V and VI) significantly. Molecular docking studies propounded that dimethoate interacted with SOD CAT and GR disrupting 3D structures and had shown strong affinity and interaction with genomic DNA. But the indirect action of dimethoate happened to be more toxic generating oxidative ROS burst destabilizing cellular homeostasis of all living root tip cells. GSE extract counterbalanced the extent and potency of ROS generation by dimethoate in A. cepa L. genome, owing to its strong polyphenolic contents. Therefore, GSE might serve as an agent of surveillance against ecotoxicological markings of different pesticides.
