Role of the Conformational State of the Promoter–Operator Region of the Bacillus cereus hlyII Gene in Interaction with HlyIIR Transcription Regulator

HlyIIR is a negative transcriptional regulator of the Bacillus cereus hemolysin II gene, binds to unusually long inverted DNA repeat (44 bp) located above the hlyII gene. It was demonstrated that the dimeric form of the HlyIIR protein binds to various regions of the hlyII gene operator region. HlyIIR is able to bind only to double-stranded DNA in the form of a mirror-symmetrical palindrome. A single HlyIIR dimer interacts with both flanking regions of the operator and its central region. The efficiency of formation a complex between the regulator and flanking sequences in the experiments in vitro is lower than for a full-size operator and its central part. The synthetic operator with a size of 50 nucleotides, which forms a hairpin due to the mirror symmetry of the oligonucleotide, does not interact with HlyIIR. Based on the obtained experimental results, it can be proposed the binding site has the following structure: TTTWAAANNNNNNTTTWAAA, where W is A or T.
