Abstract <p>Aging is an inevitable and complex process associated with organ physiological dysfunction and decreased life quality. <i>KLOTHO</i> (<i>KL</i>) is identified as an anti-ageing gene, which plays an important role in developing many age-related diseases. The objective was to analyze the methylation status of the <i>KL</i> gene in 152 subjects by using methylation-sensitive high-resolution melting (MS-HRM). To assess age-related variations in the methylation status of the <i>KL</i> gene, subjects were categorized into two groups: ≤39 and ≥40 years old. A significant association between <i>KL</i> methylation status and age was identified (<i>P</i> = 0.0009). However, this study did not determine an association between <i>KL</i> methylation status and Body Mass Index (BMI) (<i>P</i>&#xa0;&gt; 0.05). We demonstrated that α-KL methylation is increased with age in patients with ≥40 years.</p>
