Abstract <p>The World second largest man made, Asia first largest and declared as a Ramsar site “the Hirakud dam” is an important ecological barrier of the Mahanadi River basins located near Sambalpur with great diversity of fish species. However, fish from these regions are threatened by anthropogenic activities, including overfishing and exotic fish. Here, we employed DNA Barcoding to represent the fish diversity by using partial region of Cytochrome oxidase (COI) of the mitochondrial genome. We obtained 43 mitochondrial COI barcode sequences of fishes from 27 species, 8 families and 7 orders. The average Kimura two parameter (K2P) distances within species and genera were 0.36 and 9.36%, respectively. The average interspecific distance was 26 times higher than the mean intraspecific distance. The Barcode Index Number (BIN) discordance report showed that 43 specimens represented 27 BINs, of which 19 were singletons, 8 were taxonomically concordant. Present study is provided significant contribution towards the fisheries statistics of this area, which would facilitate assessment of species catch composition. It is an important input to the DNA barcode library of Indian fishes and to the global DNA barcode entries in general.</p>
