The Structure of STR Allele Pool of the Population of Kholmogory Bulls from Semen Cryobank Preserved in the Komi Republic

Microsatellite polymorphism was studied in 162 bulls, including purebred Pechora-type bulls of Kholmogory breed, Kholmogory and Holstein bulls, and Kholmogory—Holstein crossbreeds. At 15 microsatellite loci, 132 alleles were identified, or 8.8 alleles on average per locus. Of the total number of alleles, 78 (59.1%), or 5.2 alleles per locus on average, were found with the frequencies higher than 0.1 in at least one of the groups of bulls of different pedigree and full-bloodedness. Twenty-one alleles at 13 loci were found with the frequencies of 0.15 and higher, regardless of the breed, pedigree and full-bloodedness of the group. The highest number of alleles detected in the animals within one sample was revealed in the Pechora—Kholmogory—Holstein and Kholmogory—Holstein crossbreed groups. The maximum genetic distance was revealed between the Pechora—Kholmogory crossbreed bulls and Holstein bulls (DN = 0.237, FST = 0.045). High genetic differentiation of Holstein bulls with crossbreed and purebred Pechora-type Kholmogory sires was supported by cluster analysis. The genetic difference between classic Kholmogory breed and Holstein breed was not so high. Cluster analysis of the data on microsatellite genotyping of Kholmogory bulls in one array with the samples of Holstein animals and Kholmogory—Holstein crossbreeds made it possible to obtain additional information for planning measures to maintain the genetic diversity of the preserved herd of purebred Kholmogory cattle.
