Analysis of Genetic Identity of Embryogenic Cell Lines of Larix sibirica

The paper deals with the investigation of genetic identity of genotypes by 21 microsatellite loci of a donor tree, embryogenic cell line (CL6) produced from a donor tree, and clones grown from CL6 and embryogenic CL22.27.1 for which the explant (zygotic embryo) was produced as a result of controlled pollination of a clone with pollen from a maternal donor tree. Genotyping by nuclear microsatellite loci of a donor tree and CL6 showed a partial identity of detected alleles by the majority of examined loci. Variability of individual loci in the samples indicates manifestation of paternal genotype caused by introduced pollen inevitable for free pollination. Genotypes of clone needles entirely corresponded to CL6. In CL22.27.1, most loci contained alleles lacking in parental genotypes. Only two loci were identical to parental genotypes. The high frequency of mutations in the produced cell line indicates its genomic instability.
