Exopolysaccharide Production by Bacillus sp. NIOTSM16 from Arabian Sea Seamounts: Characterization and Optimization

Bacterial exopolysaccharides (EPS) have been received attention due to its potential biomolecules with significant applications. Exopolysaccharide producing marine bacterial strain NIOTSM16 was isolated from the seamounts in Arabian Sea. The strain was identified to be Bacillus species based on morphological and molecular characterizations. The identified strain NIOTSM16 produced 4.5 g/L of EPS in medium containing yeast extract, peptone supplemented with sucrose. Further, the strain was optimized for growth and EPS production under various physiological conditions. The bacterial strain was found to be facultatively alkaliphilic as it showed luxuriant growth and high EPS production at pH 11. The total amount of carbohydrates and proteins in EPS were found to be 7.9 and 0.002 mg/mL. EPS produced by strain NIOTSM16 was characterized using UV and FTIR spectrometric analyses. The UV spectrometry revealed significant absorption peak corresponding to carboxyl, carbonyl, amine and ester groups while the FTIR spectrum indicated monosaccharides with pyranosidic configuration. The chemical composition of the produced EPS through GCMS suggested the presence of glucopyranoside, ribofuranose, fructose, octanoic acid, hexadecanoic acid and octadecanoic acid. EPS produced by the bacterial strain also showed significant antioxidant capacity. The results indicated that the seamounts remain a potential hotspot for bioprospection and the EPS produced by Bacillus sp. NIOTSM16 could be evaluated for biotechnological and industrial applications.
