Abstract <p>Acute or chronic alcohol intoxication leads to significantstructural, functional, and metabolic impairments in skeletal musclefibers of mammals, including humans. Approximately 50% of chronic heavydrinkers develop chronic alcoholic myopathy, characterized by muscleweakness and atrophy in the proximal limb muscles. Muscle atrophyinduced by excessive alcohol consumption is largely driven by a reductionin muscle protein synthesis, which, in turn, results from the suppressionof various signaling pathways responsible for ribosomal translation. Thisreview summarizes recent advances in research on the effects ofacute and chronic alcohol consumption on key molecular regulatorsof protein synthesis in the skeletal muscles of laboratory animalsand humans. Furthermore, the review identifies potential directionsfor future research into the molecular mechanisms of alcohol-inducedskeletal muscle damage and discusses potential therapeutic strategiesfor the management of alcoholic myopathy.</p>
