Studying the Glutamatergic System of the Brain in PM Rat Strain with Pendulum-Like Movements and Predisposition to Audiogenic Epilepsy

The PM rat strain (an abbreviation of the words “pendulum-like”and “movements”) was obtained by selection for enhanced stereotypicalpendulum-like head movements, which is one of the forms of manifestationof catatonia. The catatonic syndrome is characterized by psychomotordysfunction and can occur in individuals with various psychopathologiesand neurological diseases, as well as with pharmacological effects.In addition to stereotypy, a predisposition to audiogenic epilepticseizures was also noted in the PM rats in the process of selection.It is known that the pathogenesis of epilepsy is associated withan imbalance of excitation and inhibition processes in the centralnervous system, in which the activity of the glutamatergic systemplays an important role. To assess the contribution of the brainglutamatergic system of PM rats to the development of catatonic-likeand epileptic symptoms, the aim of the work was to study the amountof glutamate and glutamine in the brain structures (hippocampus,frontal cortex, striatum) and blood serum and the expression ofthe brain glutamatergic system genes in PM rat strain. Using real-timePCR method, an increase in the amount of mRNA of the Grm2 and Slc1a3 genes,a decrease in the amount of mRNA of the Gad1 inthe hippocampus, as well as an increase in the amount of mRNA ofthe Grm2 and Grm5 genes in the frontal cortex,were demonstrated in PM rats. HPLC method demonstrated an increasein the amount of glutamate in the hippocampus of PM rats, whichcan affect the level of excitation in the brain and, together witha decrease in glutamine in the hippocampus and blood serum, reflectschanges in glutamate metabolism. Data obtained indicate the involvementof the glutamatergic system in the development of catatonic-like andepileptic symptoms in PM rat strain and contribute to the developmentof ideas about the hippocampus as a center of epileptogenesis.
