Applicability of Mitochondrial Gene COI for Identification of Diaspidiotus perniciosus (Comstock) (Hemiptera, Coccoidea: Diaspididae)

The data obtained provide an assessment of variability of the COI gene region for diagnostics of Diaspidiotus perniciosus (Comstock, 1881). Nucleotide sequences were obtained for the first time for five armored scale species distributed in the Palaearctic: D. ostreaeformis (Curtis, 1843), D. slavonicus (Green, 1934), D. danzigae Kuznetsov, 1976, D. zonatus (Frauenfeld, 1868), and D. caucasicus (Borchsenius, 1935). The COI gene region is characterized by a rather high degree of polymorphism and the presence of a barcoding gap, as shown by genetic distances determined within the Kimura two-parameter model. The analysis of 7 geographic populations of D. perniciosus has revealed 5 haplotypes, 4 of which are found in Russia. The results obtained are of practical significance for diagnostics of D. perniciosus by DNA barcoding.
