<p>This in vitro study aimed to evaluate the effect of four irrigation solutions [sodium hypochlorite (NaOCl), ethylenediaminetetraacetic acid (EDTA), Dual Rinse HEDP (DR-HEDP), and Triton] on the accuracy of measurements performed by three apex locators (Woodpex III Gold Plus, EndoRadar Pro, and X-Smart Pro +) using a hand file, and by the two integrated motors (EndoRadar Pro and X-Smart Pro +) using a glide path file with the ‘auto-stop (AS)’ mode. The actual working length (AWL) was determined for 240 Vertucci type 1 teeth. A total of 80 teeth were allocated for the hand file measurement groups, which were divided into four irrigation solution groups (n = 20 each). A sequential protocol was applied within each group. For instance, in the NaOCl group, the electronic working length (EWL) was measured sequentially using the Woodpex III Gold Plus, EndoRadar Pro, and X-Smart Pro + , with the solution being refreshed between each device measurement. This sequential process was repeated for the remaining three solution groups (EDTA, DR-HEDP, and Triton) (n = 20 for all 12 groups). Separately, 160 teeth were assigned for measurements in AS mode using the glide path files integrated into the two endomotors. These teeth were first divided into two main groups (n = 80 each) and subsequently subdivided into 4 groups based on the irrigation solution used, and the EWL measurements were completed (n = 20 for all 8 groups). AWL and EWL differences were evaluated within a ± 0.5&#xa0;mm tolerance. The main effects of the solution and apex locator were not significant (<i>p</i> &gt; 0.05). No significant difference was found between the hand file and AS mode (<i>p</i> &gt; 0.05). The risk of measurement values falling outside the ± 0.5&#xa0;mm range was 2.158 times higher in the NaOCl compared to the Triton (p = 0.036). Triton and DR-HEDP allowed for measurements to be within the ± 0.5&#xa0;mm range in a greater number of samples.</p>
