A comparative transcriptional atlas of the early injury response between rat (Rattus norvegicus) and pig (Sus scrofa) skeletal muscle

Comprehensive cross-species molecular datasets of tissue repair are essential for translational research but remain scarce. We present a high-resolution transcriptomic resource profiling the acute response to standardized skeletal muscle injury in rats and pigs. Tissues were collected at seven time points post-injury, including 4, 8, 12, 16, 20, 24, and 48 hours, together with an uninjured control, capturing the progression from damage through inflammation to the onset of early repair. Raw and processed RNA-seq data are publicly available in the Gene Expression Omnibus repository. Detailed quality control metrics confirm technical robustness, including per-base quality scores, GC distribution, mapping rates above 97% for rat and 96.8% for pig, and uniform gene body coverage. Example comparative analyses demonstrate the dataset’s utility, showing that temporally correlated gene modules and concordant expression trajectories are shared across species, while species-specific differences are observed in the kinetics of myogenic regulator expression. Candidate cross-species gene sets are provided as supplementary tables. This resource enables comparative analysis of conserved and species-specific repair pathways and facilitates biomarker discovery for injury staging.
