Essential oils, phytochemical and antibacterial activity of Stachys species (S. inflata and S. acerosa) collected from natural habitats in Iran

The genus Stachys has been widely utilized due to its diverse medicinal properties. This study was conducted to identify essential oil (EO) constituents, antioxidant activity, and phytochemical profiles of twelve Iranian populations from Stachys inflata (SIN) and Stachys acerosa (SAC) collected from Iran. The EO components were analyzed using gas chromatography-mass spectrometry (GC and GC–MS). A total number of forty chemical compounds, which accounted for 90.08–94.8% of the total composition of the EO were identified. These compounds belong to seven chemical groups, that oxygenated sesquiterpenes (13.66–26.05%) and sesquiterpene hydrocarbons (22.4–46.37%) were identified as two major groups in this study. Four distinct chemotypes were identified based on the EO chemical compounds. All SIN populations (SIN1, SIN2, and SIN3) exhibited separate chemotypes, with major chemical constituents including germacrene D (3.12–24.62%), spathulenol (1.07–7.68%), and β-eudesmol (2.45–2.82%). The EO of studied populations exhibited moderate antibacterial activity against both Escherichia coli and Staphylococcus aureus. In view point of phytochemical traits, the highest levels of total phenolics content (10.72 mg GAE/g DW), total flavonoids (9.09 mg QE/g DW), total flavonols (3.23 mg QE/g DW), and anthocyanins (69.57 nmol/g FW) were found in SAC8, SIN2, SAC4, and SAC7, respectively. Antioxidant activity, assessed through the DPPH assay, ranged from 68.77% in SAC7 to 86.9% in SIN1. The high diversity observed among different SIN and SAC populations provides good potential for selecting the best populations and using them in domestication projects, cultivation and integration of these species into the food and medicinal industries.
