Extraction process and novel RP-HPLC approach for polyphenols and biopotentiality of underutilized Blastania cerasiformis and B. garcinii

Novel RP-HPLC approach was developed and validated to determine ten phenolics from Blastania cerasiformis and B. garcinii. The adapted extraction method found efficient to isolate polyphenols fractions from leaves and fruits (unripened and ripened) of Blastania species, which were ensured using HPLC, LC–MS and spectrophotometric analysis. RP-HPLC noted coumaric acid, ferulic acid, vanillic acid, hydroxybenzoic acid and gallic acid as major compounds. LC–MS confirmed 12 bioactive metabolites among which 7 are phenolic acids. The spectrophotometric analysis noted highest polyphenols content [total phenolic (TPC), total flavonoid (TFC), total tannin content (TTC)] and antidiabetic activities [α-amylase inhibitory (AAIA), α-glucosidase inhibitory activity (AGIA)] from B. cerasiformis leaves. The maximum antioxidant potential was exhibited from B. garcinii ripened fruits through DPPH, FRAP and PMA assays. Besides, all tested polyphenols fractions marked for notable antimicrobial activity against Bacillus cereus and B. subtilis and Staphylococcus aureus. The extensive research needed to utilize Blastania species as novel resources for the discovery of new therapeutic agents.
