Phytochemical analysis of polyphenol-rich extract of Ocimum gratissimum leaves by GC–MS, TFC, TPC, TTC, and in vitro antioxidant properties

Ocimum gratissimum (OG) has been used as a condiment, local herb, and in the management of various ailments. Our investigation pointed out 21 compounds of best quality from the NIST14 library by GC–MS evaluation of polyphenolic extract of Ocimum gratissimum (PREOG) leaves with different activities. Qualitative analysis displayed an abundance of alkaloids, anthraquinones, flavonoids, phenols, tannins, and terpenoids. The quantities of steroids and glycosides were small, while saponins were detected. PREOG yielded 203.40 ± 27.59 mg/g garlic acid equivalents of phenolic content, 292.40 ± 77.63 mg/g catechin equivalents of flavonoids, and 262.20 ± 25.54 mg/g garlic acid equivalents of tannic content. The scavenging activity of PREOG, determined by DPPH and FRAP assays, revealed IC50 values of 39.90 µg/mL and 1280.70 µg/mL, respectively. Overall, PREOG contains a substantial quantity of flavonoids, phenols, and tannins with very powerful antioxidant characteristics, suggesting its usage for phyto-therapeutic purposes and as a source of natural antioxidants.
