Unlocking the potential: a specific focus on vesicular stomatitis virus as a promising oncolytic and immunomodulatory agent in cancer therapy

The review indicated that Oncolytic viruses (OVs) have transformed cancer therapy, with the Vesicular Stomatitis Virus (VSV) standing out for its potent anti-tumor immune response and specific oncolytic abilities. VSV, a single-stranded negative-sense RNA virus, selectively targets and destroys cancer cells, exploiting defects in the interferon type I (IFN) responses typical of tumors. Its broad tropism, rapid replication, and genetic adaptability make it a promising tool in cancer treatment. VSV's engineered modifications, such as altering the G protein for targeted delivery and combining it with immune checkpoint inhibitors, enhance its therapeutic potential. Preclinical studies show VSV's effectiveness against lung, prostate, breast, and liver cancers. Despite challenges like immune evasion, ongoing clinical trials aim to confirm VSV’s safety and efficacy, potentially establishing it as a significant advancement in oncolytic virotherapy.
