Microemulsion-Loaded Chitosan Transdermal Films of Posaconazole for Targeted Aspergillosis Therapy

Aspergillosis, a life-threatening fungal infection, presents significant treatment challenges due to limited therapeutic options and drug toxicity. Posaconazole, though effective, suffers from poor solubility & low bioavailability. This study aimed to develop a microemulsion-loaded chitosan transdermal film of Posaconazole to improve drug delivery and antifungal efficacy. The optimized microemulsion (MEP3) indicated size of the globules (20.52 nm), zeta potential (-2.77 mV) as well as encapsulated efficiency of the drug (94.81%). The developed transdermal film showed a sustained drug release and exhibited a cumulative release of 77.91% in 48-h and possessed good antifungal activity on a 29 mm zone of inhibition against Aspergillus niger. The present results indicate that the fabricated Posaconazole-chitosan film holds the potential to offer an upgrade on the control of aspergillosis by increasing the bio availability, slow release of the drug, and decrease the dosing frequency.
