Electrochemical Method for Rapid Screening of Hirschsprung’s Disease

Hirschsprung’s disease (HD) is a congenital disorder characterized by the absence of ganglion cells in the distal colon, leading to bowel dysfunction such as constipation and abdominal distension. Early and accurate diagnosis of HD is critical for timely treatment and improved patient outcomes. Conventional diagnostic approaches, including rectal biopsy and histopathological examination, while effective, are invasive, time consuming, and often require specialized facilities. Therefore, there is a pressing need for novel, non-invasive, and rapid diagnostic techniques for HD that can be applied in clinical and point-of-care settings. In this Review, we explore the potential of electrochemical sensors as a rapid screening tool for Hirschsprung’s disease. Electrochemical biosensors offer several advantages, including high sensitivity, specificity, ease of use, and the ability to provide results within minutes, making them ideal candidates for the development of diagnostic platforms for HD. The electrochemical method leverages the detection of biomarkers associated with the condition, utilizing a simple, cost-effective, and non-invasive approach. A rapid electrochemical-based diagnostic approach has also recently been attempted, where a biomarker-based discrimination of the affected area is identified in a portable-sensing device. This opens the path for developing a point-of-Need (PoN) device for detecting the pathological segment of the disease in a real time as well as obviates the need of the expertise of the pathologist. In this review, we discussed the perspectives for new rapid diagnostic strategies for HD and on the present and future direction for the development of PoN.
