Effects of sex steroids on gonadal neuropeptide Y expression and histomorphometry in the freshwater catfish, Clarias batrachus

The presence of neuropeptide Y (NPY) in fish gonad and a positive correlation between the increasing levels of sex steroids and NPY expression in ovary and testis with progression of gonadal activity (Priyadarshini and Lal 2018a, 2018b) in a seasonal breeding catfish Clarias batrachus indicates that sex steroids might play a role in the regulation of NPY expression in gonad. Therefore, the present study was designed to examine the role of sex steroids on NPY expression in the gonads of the catfish Clarias batrachus. For this, the acclimated catfish were treated with different doses of 17β-estradiol and testosterone during the mid-recrudescence phase and their effects on NPY expression in ovary and testis and histo-morphometry of the gonad were examined. Results suggested that 17β-estradiol stimulated the NPY expression in the ovary but not in the testis of the catfish whereas testosterone increased NPY expression in the ovary as well as in the testis. This study, for the first time provides experimental evidence of steroid regulation of gonadal NPY expression in fish. Further, treatment of catfish with 17β-estradiol and testosterone results in significant stimulation of oogenesis, as the number of advanced follicles in steroid-treated fish ovary increased substantially with concurrent reduction in the early-stages of follicles in comparison to the control catfish ovary. Testosterone (but not the 17β-estradiol) augments the testicular activity too as evident from the increase in the area and perimeter of seminiferous tubules and increase in the numbers of advanced germ cells after treatment indicates the involvement of testosterone in augmenting and fastening of the spermatogenic processes.
