Shilajit mitigates chemotherapeutic drug-induced erectile dysfunction via modulating HPG axis and NO/PDE5α signalling pathway

Cyclophosphamide (CPA), a globally used chemotherapeutic and immunosuppressive drug, is associated with several adverse effects and reproductive dysfunction, including erectile dysfunction. Shilajit, a revered traditional Ayurvedic medicine, possesses antioxidant, aphrodisiac, rejuvenating, and anticancer properties, suggesting its therapeutic potential against CPA induced erectile dysfunction.
ObjectiveThe present study aimed to evaluate the ameliorative potential of Shilajit supplementation against CPA induced erectile dysfunction.
MethodologyAdult male Parkes mice received a single IP injection of CPA (200 mg/kg body weight) on day one, followed by oral supplementation of Shilajit (100 or 200 mg/kg body weight) daily for 35 days.
ResultsCPA exposure induced marked structural alterations in the corpus cavernosum, including loss of smooth muscles, increased collagen deposition, oxidative stress, and reduced nitric oxide (NO) bioavailability. CPA also disrupted the HPG axis, leading to decreased testosterone levels and impaired libido. Shilajit supplementation significantly restored penile histoarchitecture by improving the SM/Collagen ratio. It also ameliorated NO production through upregulation of eNOS and nNOS and suppressing PDE5α, along with concomitant improvements in cGMP and pAkt levels. Furthermore, Shilajit restored dopamine and serotonin levels along with elevated LH, FSH, and testosterone concentrations, thereby improving sexual behaviour and fertility potential. These ameliorative effects of Shilajit were associated with enhanced antioxidant defence, evidenced by increased SOD and catalase activities.
ConclusionShilajit ameliorates CPA-induced erectile dysfunction by reducing oxidative stress, restoring NO bioavailability, and activating the NO/cGMP signalling pathway, thereby improving endothelial function and penile erection.
