<p>Standardizing any herbal formulation using scientific methodologies will enhance the quality and authenticity of the final product. The main aim of the current study is to standardize the AYUSH-64 coded polyherbal formulation, which consists of four ingredients viz., <i>Alstonia scholaris</i> (L.) R.Br., Apocynaceae, <i>Picrorhiza kurroa</i> Royle ex. Benth., Plantaginaceae, <i>Swertia chirayita</i> (Roxb.) H.Karst., Gentianaceae, and <i>Caesalpinia bonduc</i> (L.) Roxb<i>.</i> (syn: of <i>Guilandina bonduc</i> L.), Fabaceae. The formulation was standardized through organoleptic characterization, macro and microscopic evaluation, physicochemical testing, safety parameter evaluation, and thin-layer chromatography/high-performance thin-layer chromatography profiling as per Ayurvedic Pharmacopeia of India standards. Additionally, the presence of picroside-I and picroside-II markers in the formulation was quantified using high-performance liquid chromatography. The results of the experiments showed that all the ingredients were present and safety parameters were in compliance with Ayurvedic Pharmacopeia of India standards. Notably, the amount of picroside-II was found to be higher than that of picroside-I. The obtained results were discussed in detail in the present manuscript.</p> Graphical abstract <p></p>
