<p>Gastric ulcers are ulcerative and inflammatory gastrointestinal disorders that present a significant public health issue worldwide. This condition arises from an imbalance between aggressive and protective factors in the gastric mucosa, often exacerbated by excessive alcohol consumption. Flavonoids, such as troxerutin, are well-known in pharmacology for their antioxidant, anti-inflammatory, and cytoprotective properties. This study aimed to evaluate the gastroprotective effect of troxerutin on ethanol-induced gastric ulcers in mice, focusing on its role in the nitric oxide pathway, as well as its antioxidant and anti-inflammatory mechanisms. Gastric lesions were induced with ethanol (0.2&#xa0;ml, <i>p.o</i>.), followed by treatment with troxerutin (10 and 100&#xa0;mg/kg, <i>p.o</i>.), <i>N</i>-acetylcysteine, <span>l</span>-arginine, and <span>l</span>-NAME [<i>N</i>(G)-nitro-<span>l</span>-arginine]. To assess the gastroprotective effect of troxerutin, the following evaluations were performed: percentage of ulcerated area, histopathological analysis, measurement of glutathione, malondialdehyde, and myeloperoxidase, immunohistochemistry for tumor necrosis factor-alpha and inducible nitric oxide synthase, and molecular analyses. Results demonstrated that troxerutin exhibits a gastroprotective effect at 100&#xa0;mg/kg, with the possible mechanisms of action underlying this protective effect identified.</p> Graphical Abstract <p></p>
