Analysis of Inter-Root Soil Habitat and Physiological Traits in Healthy and Root Rot-Infested Custard Apple (Annona Squamosa L.) Plants

Many diseases reduce the custard apple yield and quality. Root rot is one of the most common diseases. This study aimed to investigate the changes in the inter-root soil microbial community characteristics, as well as the physicochemical properties and physiological growth indices of healthy and root rot diseased custard apple plants. Twenty-four inter-root soil samples were collected, including 12 healthy and 12 root-rotted plants, respectively, at a distance of about 20–30 cm from the main stem of the custard apple and at a depth of 25–35 cm. And fresh samples were randomly selected from these 24 fruit trees for physiological characterization. The soil organic matter and pH were low, and available nitrogen and alkali-hydrolysable nitrogen were high in the root rot plants. The diseased plants showed higher nitrogen contents in leaf, and lower total chlorophyll content and plant height. Soil enzyme activities was done to find that Peroxidase and β-1,4-Glucosidase could be a strong correlation with the onset of disease. From the fungi and bacteria community characteristics, it was found that the fungal and bacterial communities in diseased plants inter-root soil had higher richness. The Illumina MISEQ sequencing results indicated the Fusarium, Trichoderma, Bacteroidetes and Proteobacteria were enriched in the root-rotted plants significantly. The study showed that the infestation of root rot affected plant physiological feature, changed microbial communities composition, soil nutrient structure and enzyme activity. This study provides a comprehensive perspective on the relationship between diseases custard apples and inter-root soil.
