Morphological characterization and DNA barcoding of four Andrena species (Hymenoptera: Andrenidae) from the Kashmir valley, India

The study provides the first morpho-molecular characterization of four Andrena species newly recorded from the Union Territory of Jammu & Kashmir, India: Andrena (Agandrena) agilissima, Andrena (Micrandrena) fucata, Andrena (Taeniandrena) ovatula and Andrena (Zonandrena) gravida in order to address taxonomic ambiguities arising from pronounced morphological variability within the family Andrenidae. A comprehensive taxonomic assessment was conducted using 23 diagnostic morphological characters for four species of Andrena in combination with molecular analysis based on DNA barcoding (COI gene). Quantitative comparison of diagnostic characters revealed consistent interspecific differences in body size, pubescence pattern and clypeal sculpture, allowing reliable separation of taxa. Molecular analysis showed clear species delimitation, with interspecific genetic divergence exceeding typical intraspecific variation, confirming the distinct taxonomic status of all four species. Additionally, barcode sequences were successfully generated for all four species and deposited in Gene Bank at National Center for Biotechnology Information (NCBI). The integration of morphological and molecular data enhances the reliability of species identification within the genus Andrena. Future studies should expand geographic sampling across the Himalayan region, incorporate additional nuclear markers to resolve deeper phylogenetic relationships and develop regional identification keys to facilitate accurate biodiversity assessment and pollinator conservation planning.
