<p>The comparative analysis of araneofauna (spider populations) between Perumpalam Island (inhabitant) and Pathiramanal Island (non-inhabitant island) offers significant insights into the diversity and abundance of spiders across various ecological settings. The study reveals notable variations in abundance and diversity among sampled areas, indicating the potential influence of human presence on arachnid communities. Comparatively, intermediate disturbance areas (IDA) reported lower Richness (54 species) and diversity (Shannon = 33.84; Simpson = 24.90), while highly disturbed areas (HDA) exhibited the lowest values (28 species; Shannon = 20.83; Simpson = 16.99). PE recorded significant abundance (700 individuals) and diversity (Shannon = 37.51; Simpson = 29.11), suggesting strong influences of synanthropic species. ANOVA results indicated significant differences in spider abundance across sites (F<sub>3,44</sub> = 16.414, <i>p</i> &lt; 0.001), with LDA showing the highest mean abundance (95.75 ± 43.95). Species Richness estimation through rarefaction confirmed completeness for most habitats, with LDA at 99.4% and other areas at 100%. Notably, LDA and IDA exhibited 97.6% similarity in assemblage composition, while lower similarity was observed between LDA and PE (57.2%). Indicator species analysis identified <i>Tylorida ventralis</i> and <i>Heteropoda venatoria</i> as key species driving dissimilarities. Higher spider density and diversity were associated with habitats under low disturbance, while PE showed comparable density to IDA, influenced by synanthropic species (62%). The study highlights the impact of human activities on spider communities, with implications for biodiversity conservation on these islands.</p>
