<p>The synthetic seed technique is a viable biotechnological alternative for the mass propagation of plant species with economic value and ornamental potential. However, this technique faces challenges such as embryo mortality and variation in the efficiency of encapsulation matrices. Among ornamental plants, <i>Kalanchoe daigremontiana</i> stands out, which also has scientific value as an excellent plant model for assays. Therefore, this study aimed to evaluate different compositions of synthetic seed encapsulation matrices associated with varying concentrations of growth regulators. For this purpose, <i>K. daigremontiana</i> embryos were used as explants to assess their development. The plants were commercially obtained, and from them, embryos were extracted for disinfection. The embryos were treated with 70% ethanol for 1&#xa0;min, followed by immersion in 2.5% commercial sodium hypochlorite for 15&#xa0;min, and subsequently inoculated into the encapsulation medium. Fourteen treatments with different concentrations of agar, MS medium, and growth regulators, with or without complexation in sodium alginate and calcium chloride, were tested. Capsule composition and integrity characteristics, as well as the development of <i>K. daigremontiana</i> explants, were evaluated. The results showed that the combination of 9&#xa0;g L<sup>− 1</sup> of agar and 1/4 strength MS medium, with complexation, regardless of the use of growth regulators, resulted in a more flexible matrix, allowing greater capsule integrity and aerial part development.</p>
