Morpho-anatomical characterization with special reference to the secondary metabolites and bioactivities of Nervilia plicata (Andr.) Schltr.

This study aimed to provides information related to Nervilia plicata (Andr.) Schltr. of macro- and micro-morphology, chemical constitutes, biological activities and its medicinal uses. Macromorphologically, N. plicata is characterized by tuber whitish, globose to ellipsoid, leaves almost orbicular with broad cordate base, many veined, pubescent, petiole erect, floral bracts cuneate, acuminate, flowers nodding, widely opening, sepals and petals similar, light pale brownish-yellow, lanceolate, petals little shorter. Micromorphologically, the anatomical features of leaf, pseudopetiole, tuber, and root described in detail. Needle-shaped calcium oxalate crystals were found in the mixed leaf and pseudopetiolate powder, and mixed tuber and root powder. Oval-shaped starch grainules were found only in the mixed tuber and root powder. The in vitro antioxidant and antidiabetic activities of whole plant (except flowers) extracts of N. plicata were, anti-hyperglycemic effect of the extract was investigated by oral glucose tolerance test (OGTT) in mice. Total polyphenol content in the 100% ethanol, 50% ethanol, and water extracts was 19.94, 16.94, and 17.75 mg GAE/g d.w., respectively. Total flavonoid content in the extracts was 7.12, 8.67, and 5.16 mg QE/g d.w., respectively. In vitro activities, the 50% ethanol extract had the best DPPH radical scavenging activity and reducing power with IC50/EC50 value of 55.91 and 66.77 µg/mL, respectively. The in vitro antidiabetic activity of the extracts is very low compared to the standard used, in which, the IC50 value of the 50% ethanol extract for PTP1B inhibition was 6.011 µg/mL. The 50% ethanol extract had great anti-hyperglycemic effect at 5 and 10 mg/kg in OGTT. Overall, the N. plicata could become a promising subject for further research in the future.
