Molecular evidence of cherry leaf roll virus isolate from the phylogenetic group D1 infecting black elderberry (Sambucus nigra) in Ukraine

The majority of sequenced to date isolates of cherry leaf roll virus (CLRV) from elderberry plants belong to the phylogenetic group E, while a few CLRV isolates are also clustered into the clades A and B. In this study, CLRV was detected by DAS-ELISA and RT-PCR in Sambucus nigra leaves exhibiting vein clearing and mild mottling with no leaf rolling. Obtained sequence (409 nt) of 3′-untranslated region (3′UTR) of CLRV isolate was named Eld28Lubny-Ukr and submitted to the NCBI GenBank under the accession number OQ627304. Phylogenetic analysis showed that Eld28Lubny-Ukr belonged to group D1, and its nucleotide and amino acid identities with D1 isolates of CLRV were 96.6–99.2% and 93.6–99.0%, respectively. It is known that group D1 includes CLRV isolates mostly from Juglans regia, Betula sp. with no reports of isolates from elderberry. To our knowledge, this is the first report of CLRV isolate recovered from elderberry and belonging to the phylogenetic group D1.
