<p>Bacterial leaf spot (BLS) caused by <i>Xanthomonas citri</i> pv. <i>viticola</i> (<i>Xcv</i>), is a significant disease affecting grape production worldwide. Climatic pattern plays an essential role in the incidence of BLS. Meteorological variables strongly affects the stage of growth, host susceptibility, succulence, vigor, survival rate, multiplication ability, dispersion of pathogen, spore penetration and germination. Anticipating how these environmental factors are influencing these traits, could help in implementation of strategies for effective BLS management. Study was conducted to investigate factors affecting disease incidence and its management using <i>Bacillus</i> isolates. It was conducted at six different locations viz. Pune, Sangli, Solapur, Nashik, Vijayapur and Theni during 2022–23 and 2023–24. Results displayed that optimum values of environmental variables, i.e. temperature (28–32°C), relative humidity (23–91%) and rainfall (5 mm) were most favorable for the BLS development. It was observed that <i>B. australimaris</i> manifested superior multiplication and defense enzymes induction ability than <i>B. zhangzhouensis</i> both in presence or absence of <i>Xcv</i>. It was also reported that preventive application of <i>Bacillus</i> spp. is recommended than curative. Two <i>Bacillus</i> spp. which were screened have the ability to reduce disease severity at all the tested locations. Promising results were manifested by <i>B. zhangzhouensis</i>, alone and in combination with <i>B. australimaris</i>, on field for both the years. Leaves treated with <i>Bacillus</i> spp. exhibited increased level of peroxidase chitinase, polyphenol oxidase and β 1,3-glucanase activities when compared with untreated. Findings in current study revealed the biocontrol abilities of isolated endophytic <i>Bacillus</i> species as an excellent option to be used by agriculture sectors to have sustainable environment.</p>
