<p>The high degree of specificity between <i>Podosphaera xanthii</i> and melon results in the emergence of several physiological races, hampering the development of cultivars that remain resistant to this fungal pathogen in the long term. This study aimed to characterize the variability of <i>P. xanthii</i> populations in melon-producing regions in Rio Grande do Norte State, Brazil. The sample comprised monosporic isolates obtained from melon crops grown in a protected environment or under field conditions. Experiments were conducted in a greenhouse between 2015 and 2018. Isolates were inoculated into the following 10 differential melon lines: ‘HBJ’, ‘Védrantais’, ‘PMR 45’, ‘PMR 5’, ‘PMR 6’, ‘WMR 29’, ‘Edisto 47’, ‘MR 1’, ‘PI 414723’, and ‘PI 313970’. It was possible to identify isolate races based on the resistance/susceptibility responses of melon lines. Races 1, 2Fr, 3.5, 5, Br01, Br02, Br03, Br04, Br05, and Br06 were identified. The races most frequently associated with powdery mildew in melon crops were 3.5 and 5. This is the first record of race 3.5 in Brazil.</p>
