<p><i>Cissus quadrangularis</i>, commonly known as bonesetter, is a worthy medicinal plant in the Vitaceae family, esteemed for its fleshy, quadrangular, and green photosynthetic stem, renowned for treating musculoskeletal disorders. In September 2022, moderate to severe anthracnose symptoms were observed on the stems at the University of Agricultural Sciences, GKVK, Bangalore, and surrounding areas. The associated fungus was isolated, purified and identified as <i>Colletotrichum plurivorum</i> by employing micro-morphological characterization and targeted sequencing of multi-genomic loci namely, ITS, <i>gapdh</i> and <i>his3</i> loci and phylogenetic analysis. The pathogenicity of this species on its original host was confirmed through the fulfilment of Koch’s postulates. Further, the cultural and physiological characteristics of the pathogen was studied by evaluating how different solid media, temperature regimes and different levels of pH affect the growth and sporulation of the fungus. <i>Colletotrichum plurivorum</i> infecting bonesetter plant is found to better grow and sporulate on potato carrot agar (PCA) at 30 <sup>o</sup>C and pH 6. The findings provide significant insights on <i>C. plurivorum</i> as a novel fungal pathogen affecting <i>C. quadrangularis</i> in India and provide useful information to further develop effective management strategies.</p>
