<p>Cassava (<i>Manihot esculenta</i> L.) production is influenced by biotic and abiotic factors contributing to yield decline. Anthracnose is an important cassava disease that is caused by several <i>Colletotrichum</i> species. Varietal resistance is considered the most durable approach for cassava anthracnose. However, the reaction of cassava cultivars grown in the Philippines to anthracnose caused by <i>Colletotrichum</i> species is yet to be determined due to the lack of disease and pathogen reports. This study identified the causative agents of severe circular necrotic spots by employing combined morphological, cultural, pathogenicity, and molecular characterization using ITS, TUB2, GAPDH, ACT, CHS-1 genes. Here, <i>C. fructicola</i>, <i>C. siamense</i>, and <i>C. tropicale</i> were associated with the observed disease in the field. These pathogens were then inoculated to cassava cultivar ‘Binulak’ in a detached leaf assays and an in vivo assay. Cassava cv. ‘Binulak’ was susceptible to the three <i>Colletotrichum</i> species, showing anthracnose symptoms. This is the first confirmed scientific report of <i>C. fructicola</i>, <i>C. siamense</i>, and <i>C. tropicale</i> as cassava leaf anthracnose pathogens in the Philippines. The susceptibility of cassava cv. ‘Binulak’ to anthracnose indicates a need for selection and breeding for anthracnose resistance.</p>
