<p>Clustered Regularly Interspaced Short Palindromic Repeats (CRISPR) technology represents a transformative advancement in molecular biology, enabling precise nucleic acid recognition, cleavage, and genome editing. The elucidation of CRISPR-associated protein (Cas) mechanisms has catalyzed the development of innovative molecular detection and diagnostic platforms. Recent advancements have further expanded the applications of CRISPR through integration with biosensing technologies and portable devices, revolutionizing analytical chemistry methodologies. This review comprehensively introduces the biological mechanisms and classification of CRISPR-Cas systems, with particular emphasis on their analytical applications in molecular diagnostics, biosensor development, and miniaturized device integration. The current state-of-the-art CRISPR-based analytical platforms are also systematically discussed, highlighting their operational principles and implementation strategies. Furthermore, the review critically evaluates existing challenges and future perspectives, providing valuable insights for analytical chemists to seek to implement CRISPR-Cas technologies in diverse applications, ranging from clinical diagnostics to environmental monitoring.</p>
