First report of papaya leaf curl China virus infecting Phaseolus vulgaris in China

Phaseolus vulgaris L., also known as common bean, is one of the important cash crops in China. In this study, three P. vulgaris samples with typical leaf shrinking and yellowing were collected from Chongqing of China in 2021. Full length genome amplification revealed that all the three isolates CQ1, CQ2 and CQ3 were of 2735 nt in length, shared above 99.96% nucleotide sequence identity with each other and exhibited the highest sequence identity (92.4%~99.6%) with different isolates of papaya leaf curl Chinese virus (PaLCuCNV). Recombination analysis revealed that PaLCuCNV CQ1 originated by recombination between PaLCuCNV and tomato yellow leaf curl Viet Nam virus (TYLCVNV), and its C4 gene and corresponding protein sequence shared the highest identity with TYLCVNV. PVX-mediated gene expression indicated C4 gene of CQ1 was more virulent than that of canonical PaLCuCNV in the early stage of virus infection. This is the first report of recombined PaLCuCNV infecting P. vulgaris and these findings offer a new basis for formulating comprehensive and efficient strategies to control viral diseases on P. vulgaris.
