Molecular identification of fall Armyworm, Spodoptera frugiperda (Lepidoptera: Noctuidae) strains, through exuviae- based DNA extraction

Traditional DNA barcoding methods in entomology often rely on destructive sampling, posing ethical challenges, particularly in ecological and conservation research. This study aimed to develop and optimize a non-lethal DNA extraction technique using exuviae from fall armyworm (Spodoptera frugiperda). Exuviae, especially from the pre- pupal stage, provide a non- invasive and readily available source of genetic material. Samples were collected from S. frugiperda populations across seven locations representing different provinces. DNA was extracted using the Animal and Fungi DNA preparation Kit, followed by PCR amplification and sequencing of the COI gen. the resulting sequences were analyzed using BLAST for species and strain identification. Phylogenetic analysis of the cytochrome oxidase 1 (COI) gene successfully differentiated between the rice (R) and corn (C) strains of S. frugiperda, with two distinct genetic clusters observed. Fresher and well-preserved exuviae can be serve as a reliable, ethical, and effective alternative to traditional DNA sampling methods. The approach supports sustainable pest surveillance, genetic diversity assessment, and integrated pest management (IPM), contributing a valuable tool for non- destructive insect molecular diagnostics.
