Histopathological study of soybean (Glycine max (L.) Merr) infected by Aphelenchoides pseudobesseyi (Nematoda: Aphelenchoididae)

Aphelenchoides pseudobesseyi, previously identified as A. besseyi, is the etiological agent of Green Stem and Foliar Retention (GSFR) syndrome, which causes significant yield losses in soybean crops in Brazil. The changes this nematode induces in soybean plants throughout the crop’s growth cycle remain unclear. In this study, we have employed histological methods to investigate the parasitic process of A. pseudobesseyi in soybeans, aiming to identify and understand the key events that occur during pathogenesis. A greenhouse assay was carried out to replicate GSFR symptoms, allowing for the subsequent collection of samples for histopathological analysis. Nematodes preferred feeding on shoot apex, leaves, and flower buds. Cell collapse was present in the meristematic tissues of the shoot apex, as well as in the leaf mesophyll tissues and the reproductive tissues of flowers. These cell collapses led to the necrosis of the apical bud, the formation of angular necrotic spots on the leaves, and flower abortion, respectively. Forty-eight hours after A. pseudobesseyi inoculation, the nematode migrated externally from the inoculation sites towards the infection sites, where they grouped in higher amounts external to the stem when compared to those found within it. Nematodes also caused infection on soybean pods, but no nematodes were found on the seeds. This observation provides strong evidence that A. pseudobesseyi is unable to infect soybean seeds.
