Rapid and visual detection of the strawberries pathogenic fungi Fusarium oxysporum and Colletotrichum siamense by one-pot LAMP-CRISPR/Cas12b

Soil-borne fungi such as Fusarium oxysporum and Colletotrichum siamense may lead to significant decline in strawberry production. Therefore, reliable and fast detection methods are urgently needed. Here we introduce a one-pot isothermal detection method integrating loop-mediated isothermal amplification (LAMP) and CRISPR/Cas12b. The method shows high pathogen specificity, with a detection limit of 10 DNA copies. Pathogen detection is completed within 30 min, which does not require specialized laboratory equipment and greatly increases the feasibility of rapid field testing. The establishment of this method provides a novel technical means for early warning and control of strawberry diseases and may contribute to reduction yield losses.
